You can easily see and understand both sides of the conflict here. The guard wanted to protect his city. Isaac wanted to pass through on his way to his ultimate location. Isaac offered a peaceful solution, the guy rejected, not trusting the monsters. Fair.
The guard tried using force and intimidation to get his way. Basically threatened Isaac's life. Isaac took it for the threat that it was, countered and proved his forces were stronger.

The guard mentions that they KNOW that there are demons/night creatures rampaging accross Wallachia. The sheer fact that he's telling them to get back on their ship and leave is more than a chance ANY other human had ever given Isaac. If he had just thought for even a second, he would have realized that even if the guard captain let him pass, those kind of things going throught he streets, even escorted, would cause chaos to break out. He's trying to stop it all. He may think they're evil, disgusting monsters but he knows that attacking outright will spill blood and simply allowing a master of demons to walk through will also spill blood, he says the only thing that makes sense. "DONT COME INTO GENOA." Isaac is just still pissed off at humanjty here and cant let go of his hate to remember that they're protecting people just like he wanted to protect Dracula.

Isaac was a super cool character and fascinating to follow, but I can't pretend that guard wasn't 300% justified to react the way he did. By that point though, Isaac was so done with humans he couldn't stop and see it from his point of view - unlike many who were rude to Isaac because they were assholes and even wanted to hurt him, this man had something to protect.
