Charkov knew he was poisoned when he drank the tea and put the card on the table, knowing Belikov wanted to get the card as his Closest friend, he destroyed the glass, removing the evidence. Charkov always knew Dimitri was the mole. When he accused you of being the mole and you drank the wrong cup, he still was caring for you, asking what's wrong (Dimitri, what's wrong?), this shows how close they are as friends. For me, this felt like a sacrifice more than a betrayal. Heartbreaking.
Edit : He also did toss the card out when you poisoned yourself, and when he got poisoned, he didn't take the card or anything, he just destroyed the evidence.
