A time-lapse apoptosis assay was performed with Jurkat cells to demonstrate how the CellASIC ONIX M04T trap plate can be used to track non-adherent mammalian cells over time. Jurkat cells were loaded onto the M04T plate and then perfused for 2 hrs with RPMI-1640 media containing BioTracker NucView® 488 Green Caspase-3 Dye (labels the middle stage of apoptosis) and Annexin V Texas Red (labels the late stage of apoptosis). Cells were then perfused with media containing 2.5 µg/mL CD-95 antibody, an inducer of apoptosis. Time-lapse imaging was performed at 10-minute intervals using FITC & Texas-Red filters. As five trapped cells were tracked during the apoptosis experiment, the varying temporal response to the apoptosis inducer can be observed supporting the need for single cell observation tools.
