📍 Methadone is a synthetic opioid. It is an opioid receptor agonist, NMDA receptor antagonist, norepinephrine and serotonin reuptake inhibitor. Common side effects of methadone include: constipation, loss of appetite, dry mouth, dyspepsia, and respiratory depression.
📍 Methadone has less neurotoxicity than morphine due to lack of active metabolites. Methadone is a major CYP3A4 substrate, so, avoid use with inhibitors or lower methadone dose. This drug can decease testosterone level and contribute to sexual dysfunction. Lastly, Methadone can prolong the QT interval, possibly by interacting with cardiac potassium channels. Baseline and routine ECG monitoring is recommended.
